Elucidation of dynamic interactions of histone tails with Nucleosomal or linker DNA
The histone tails protruding from the nucleosome core particle are highly flexible and are not visible in X-ray crystallography or cryo-electron microscopy structures. Also, the linker DNA that connects nucleosomes is dynamic unless the linker histone H1 is bound. Although the histone tails are dynamic, its movement restricted by the linker DNA and the nucleosomal DNA. In this study, we will focus on histones H3 and H4, which are highly conserved among species, and use solution NMR to reveal changes in the dynamic interactions between histone tails and linker DNA in heterochromatic nucleosomes. The purpose of this study is to elucidate the importance of the dynamic interaction of histone tails through DNA.
